Suppression of Intestinal Polyposis in Apc Mice by Inhibiting Nitric Oxide Production
نویسندگان
چکیده
Inducible nitric oxide synthase (iNOS) was shown to be expressed in normal mucosa and adenoma of small and large intestines of Apc mice by reverse transcription-PCR and immunohistochemistry. Administration of the iNOS inhibitor aminoguanidine (1.5 g/liter) in drinking water or an L-arginine-deficient diet to Apc mice resulted in a significant decrease in adenoma development in the small but not the large intestine. Similarly, iNOS-gene knockout Apc mice (Apc iNOS / or Apc iNOS / ) developed significantly fewer adenomas in both small and large intestines than Apc iNOS / mice. These results suggest that iNOS-selective inhibitors could be used as a potential chemopreventive agent for colorectal cancers.

متن کاملChronic epithelial NF-κB activation accelerates APC loss and intestinal tumor initiation through iNOS up-regulation.
The role of NF-κB activation in tumor initiation has not been thoroughly investigated. We generated Ikkβ(EE)(IEC) transgenic mice expressing constitutively active IκB kinase β (IKKβ) in intestinal epithelial cells (IECs). Despite absence of destructive colonic inflammation, Ikkβ(EE)(IEC) mice developed intestinal tumors after a long latency. However, when crossed to mice with IEC-specific allel...
متن کاملImpact of dietary amino acids and polyamines on intestinal carcinogenesis and chemoprevention in mouse models.
Colon cancer in humans is influenced by both genetic and dietary risk factors. The majority of colon cancers have somatic mutations in the APC (adenomatous polyposis coli) tumour-suppressor gene. Dietary arginine enhances the risk of APC-dependent colon carcinogenesis in mouse models by a mechanism involving NOS2 (nitric oxide synthase 2), as elimination of NOS2 alleles suppresses this phenotyp...
متن کاملSuppressive effects of the NADPH oxidase inhibitor apocynin on intestinal tumorigenesis in obese KK‐A y and Apc mutant Min mice
Obesity is a risk factor for colorectal cancer. The accumulation of abdominal fat tissue causes abundant reactive oxygen species production through the activation of NADPH oxidase due to excessive insulin stimulation. The enzyme NADPH oxidase catalyzes the production of reactive oxygen species and evokes the initiation and progression of tumorigenesis. Apocynin is an NADPH oxidase inhibitor tha...
متن کاملSynergistic Effect of LPS, IFN- and Iron on Apoptosis of Balb/c Mice Macrophages Following Nitric Oxide Production
Objective(s) Previous studies have demonstrated that the nitric oxide (NO) dependent death of murine peritoneal macrophages activated in vitro with IFN-g and LPS is mediated through apoptosis. In the present study, we investigated the synergistic effect of LPS, IFN-g and iron on NO production and apoptosis. Materials and Methods After determination of iron cytotoxicity, the peritoneal macrop...
